Research Abstract |
In present research, action mechanism and active ingredient of a Kampo (Japanese herbal) medicine "Shoseiryuto (SST)" against airway infection have been investigated. 1. Effect of SST against airway inflammation Oral administration of SST reduced the airway hyperresposiveness and ovalbumin (OVA)-specific IgE antibody titer in bronchoalveolar lavage fluid (BALF) of OVA-sensitized allergic airway inflammation model mice. SST also recovered the amount of appearance of spectrin a2, a cytoskeletal protein, in lung tissue of the model mice. However prednisolone, a steroidal drug, did not reduce the OVA-specific IgE Ab titer in BALF and did not recover the amount of appearance of spectrin a2 in lung tissue of the model mice. These results indicate that action mechanism of SST is different from that of prednisolone against airway inflammation. 2. Adjuvant active substance for nasal influenza vaccine from SST It has been appeared that SST shows a potent oral adjuvant activity for nasal influenza vaccine. We performed the isolation and characterization of an orally active adjuvant substance from the tuber of Pinellia ternata, as an active herbal component of SST. The active compound was identified as 9, 12, 13-trihydroxy-10E-octadecenoic acid (pinellic acid). However, stereoisomers of pinellic acid did not show the adjuvant activity individually, and combination of 9S, 12S, 13S and 9S, 12R, 13R stereoisomers (90.4 : 9.6) showed a potent activity. The combination with the nasal vaccine also protected the influenza virus infection. This is a first report of the combination effect of stereoisomers on the pharmacological activity of Kampo medicines. These results suggest that SST shows its pharmacological activities through airway mucosal immune system and pinellic acid is one of active substances in SST for airway mucosal immune system.
