Research Abstract |
We investigate the role of CYP2J2 and soluble epoxide hydrolase and EET in the cardiovascular system. In patients with type II diabetes mellitus, single nucleoetide polymorphism of soluble epoxide hydrolase is associated with progression of insulin sensitivity. Fish oil, eicosapentaic acid increased the mRNA level of CYP2J2 which synthesize EET by activation PI-3 kinase and AKT in vascular endothelial cell. This result suggests that EET production by intake of fish oil might be involved in the mechanism of vascular protective effect of eicosapentaic acid. Several drugs including angiotensin receptor blocker also increase the level of CYP2J2 and EET production. This result may add the new mechanism of angiotensin receptor blocker on vascular endothelial function. Further study of angiogenesis by EET will be needed for the treatment of severe atherosclerosis. We proposed the new disease criteria "Vascular failure". Vascular failure include both endothelial dysfunction and smooth muscle dysfunction. Depletion of EET may promote the endothelial failure and EET is one of candidate of drug for vascular failure.
