Research Abstract |
1) Extraction of total RNA from the preserved pancreatic tissue of type 1 diabetic patients and expression of inflammatory cytokines We extracted total RNA from the pancreatic tissue of type 1 diabetic patient that was taken by laparoscopic pancreas biopsy and have been preserved at-80C for several years. They were 6 patients (3 patients with fulminant diabetes and 3 patients with autoimmune type). We generated cDNA from the total RNA taken as above and examined the expression of pancreatic hormones and inflammatory cytokines by using semi-quantitive PCR methods. First of all, we confirmed the expression of insulin and glucagon gene by using these methods. Next, interferon-gamma was expressed in the pancreatic tissue of all 6 patients (3 patients with fulminant diabetes and 3 patients with autoimmune type). Tumor necrosis factor alpha was expressed in all 3 patients with fulminant diabetes but not in 3 patients with autoimmune type at all. These findings suggest the different profile of gene expression in the pancreatic tissue between fulminant and autoimmune types even if both of them are categorized within type 1 diabetes. 2) Comprehensive analysis of the gene expression by using microarray On the basis of the above findings, we tried comprehensive analysis of the gene expression in the pancreatic tissue of type 1 diabetic patients by using microarray, but failed.
