Research Abstract

1. Clinical sample from cancer patient-Twenty samples of surgical specimens from esophageal cancer patient were successfully collected for RNA analysis and primary culture.
2. Dermokine expression assay---Real-time PCR assay show that expression levels of dermokine-beta were low in cancer tissue.
3. Primary culture of resected esophageal cancer tissue---We add recombinant dermokine-beta to cultured primary cell to clarify the function of dermokine-beta in squamous cell carcinoma cell.
4. Large scale purification of dermokine-alpha, beta---We successfully established 293EBNA-1 cell expression system to generate recombinant dermokine-beta protein. Large scale purification of dermokine-alpha were not successful.
5. Variant isoform of dermokine-We analyzed morphological change of esophageal cancer cell under various concentration of recombinant dermokine-beta, but we could not find obvious changes. Recently various isoform of dermokine were reported in other group. Now we focused on isoform specific function of dermokine in cancer cell.
