Research Abstract |
The osteoporosis which combines in rheumatoid arthritis brings about bone fragility, lumbago and backache. Therefore, it is the serious complication which existing ADL failure is exacerbated. The effects of drug therapy for osteoporosis of the collagen-induced arthritis (CIA) rat which is animal model of RA, the relationship between knee joint deformity in human RA and osteoporosis, and influence of glucocorticoid (GC) on bone were investigated. 1. The inhibitory mechanism of bone resorption by minodronic acid, a nitrogen-containing bisphosphonate. in C1A rats was investigated. Minodronic acid inhibited the differentiation and the activation of osteoclasts not by inducing apoptosis but by inhibiting the RANKL-RANK, system, and thereby suppressing bone resorption. 2. The effects of COX2 inhibitor (celecoxib) on arthritis and bone mineral density in CIA rats was investigated. The influence of estrogen therapy on changes in arthritis and bone mineral density was evaluated using an estrogen-replete CIA rat model. Estrogen could suppress arthritis and bone loss in estrogen-replete rats. This finding indicates the potential usefuleness of estrogen as an effective treatment for pre-menopausal RA patients as well as post-menopausal ones. 4. The relationship between knee deformity and osteoporosis in women with RA was examined. Knee deformity in RA was derived from deformity of the proximal tibia, and it was significantly correlated with age and bone mineral density in the lumbar spine and radius. Therefore, it is considered that preservation of the systemic bone mineral density is necessary for prevention of knee deformity in. RA. 5. The influence of GC on bone in rats at different age was investigated. Six-month-old rats treated with 20 mg/kg prednisolone showed dominant cancellous bone decrease and reduced bone turnover. Less
