Research Abstract |
For mouse hepatitis virus pollution of university animal center in spring of 2006, all our mice were killed and the animal experiments were ceased. Since virus infection was re-appeared half a year after disinfection, however, the animal experiment center has been closed yet. We, therefore, had to switch plans of this study. (1) Analysis of mouse endometrial remodeling process and identification of murine endometrial stem cells: Short-term endometrial rebuilding was succeeded in by mixed culture transplant of plural endometrial cell strains, but animal experiment is stopped by the above reason, and long-term observation cannot be finished. (2) Analysis of human endometrial remodeling process and identification of human endometrial stem cells: <1> Cellular characterization of human endometrial stromal cells (i) Influence of various bioactive substances: Influence such as various cytokines, bioactive substances, cell death inducers such as anticancer drugs, siRNA experiments were examined (pr

eparation in submission). Involvement of TRAIL and TRAIL receptor was made clear by DAPK knockdown experiments (submitted). (ii) Apoptosis sensitivity: Endometrial stem-like cells had resistance to various apoptotic signals such as anticancer drugs and Fas stimulants. (iii) Differentiation activity: Partial differentiation was confirmed by mixed culture experiments. <2> Establishment of in vitro differentiation of human endometrial stem-like cells to make differentiation pedigree of human endometrial cells Partial differentiation phenomenon was confirmed by mixed culture experiments, but the experiments are ongoing now. Establishment of endometrial reproduction medical treatment by mouse intrauterine transplantation of two human endometrial stem-like cells. Animal experiments have been stopped because of the above reasons. Three human endometrial stem-like cell-specific antigen were identified. In order to establish hybridomas producing anti-endometrial stem cell monoclonal antibodies balb/c mice were immunized with human endometrial stem-like cells. However, immunized mice were killed due to contamination of the animal center. When the animal experiments were allowed, the plan will be re-started. Less
