Research Abstract |
1. Aim : To investigate the effect of inflammatory cytokine (IL-1β, IL-6) for intestinal motility, we simultaneously measured intracellular calcium concentration and tension in guinea pig taenia coli and Hirschsprung disease model rat colon. 2. Materials & methods : The residual longitudinal muscle tissue preparation was cut into strips (1×5×0.5 mm). the guinea-pig taenia coli strips were loaded with Ca^<2+> indicator dye, fura-2 in the form of acetoxymethyl ester (Fura-2/AM). The changes in the fluorescence intensity of Ca^<2+> binding with fura-2 loaded taenia coli strips were simultaneously monitored with a front-surface fluoromater CAM-OF3 (Japan Spectroscope. Co, Tokyo, Japan). 3. Results : IL-1β and IL-6 are not effective for the contraction induced by calbachol in guinea pig taenia coli. The intracellular calcium concentration elevation was not observed. These result was due to the disorder of the smooth muscle cells with IL-16 and IL-6. The strips might be incubated in IL-16 and IL-6 too much time. On the other hand, We studied about the motility of the Hirschsprung disease model rat intestinal smooth muscle. In the aganglia intestinal smooth muscle, the elevation of contraction induced by substance P was lower than in the normal intestinal smooth muscle. This result is assumed to be due to the depression of the calcium channel on the aganglia smooth muscle cell membrane. To investigate the Rho kinase pathway in aganglia intestinal smooth muscle, we used Rho kinase inhibitor (Y-27632). In aganglia intestine, Rho kinase pathway is normal. It is not assumed that Rho kinase pathway is the cause of intestinal disorder in Hirschsprung disease.
