Research Abstract |
For in situ hybridization, skull tissues in 4 week-and 40 week-old Wistar rats were fixed in 4% paraformaldehyde solution by using microwave treatment. Results 1: In 4 week- and 40 week -old normal Wistar rats, spindle cells around blood vessels in mesenchymal tissues expressed BMP-2 and-4 receptor mRNA. Preosteoblasts and osteoblasts expressed osteopontin mRNA. Osteoblasts and osteocytes expressed osteocalcin mRNA. Results 2: In the elevated and repositioned periosteum of 40 week-old Wistar rats, the periosteal structure was unclear and spindle cells expressed BMP-2 and -4 receptor mRNA. Osteopontin mRNA was expressed in spindle cells (pericapilary cells) and preosteoblasts. According to proceeding of the periosteal regeneration, osteopontin mRNA was strongly expressed in preosteoblasts and osteoblasts. Osteocalcin mRNA was first found in osteoblasts and osteocytes in the calcifying area. Results 3 : For RNA analyses, Osteopontin mRNA was strongly expressed, but osteocalcin mRNA was not detected in the repositioned periosteum.
