Research Abstract

サカナ、ヤツメウナギとカエル(Xenopus)についてToll-like receptor(微生物成分のパターン認識レセプター)のファミリーを同定した。これらをサカナ細胞などに発現させて強制発現とRNAi knockdownにより、機能査定を行なった。サカナ、カエルにあってトリ、マウス、ヒトにないTLR遺伝子,TLR14,TLR21,TLR22,TLR23を同定できた。これらは陸生の脊椎動物に無いことから水棲微生物の特異パターンを認識すると云う仮説を立てて認識微生物のパターンを検索した。TLR22はRNA認識に関与するTLRであった。しかし、ヒトのRNAパターン認識TLRとは相同性がなく、認識分子の性質も異なっていた。RNA認識のヒトTLR orthologs,TLR7,TLR3はこれとは別に存在していた。サカナTLR3も2重鎖RNAを認識するのでサカナを含む水棲生物は水棲のRNAウイルスに対応してこのTLR22系を保存させたと考えられた。
TLR21はlipoproteinを特異認識した。ヒトTLR2 subfamilyはlipoproteinを認識するが、peptidoglycanも同時に認識する。TLR21はpeptidoglycanを認識せずにVibrio菌類のパターンとしてlipoproteinを特異認識するらしい。これも水棲に適応した結果であろう。これらの示唆する所はTLR系は微生物環境に適応して構成的に認識パターンを変えることである。獲得免疫にIgでなくLRRモチーフを使っているヤツメウナギでも水棲TLRが存在するので、TLRの微生物認識は獲得免疫の発達とは独立して必要であったと考えられる。即ち、TLR依存性の免疫細胞刺激は微生物(=非自己)認識だけの問題ではなく、シグナルによる免疫担当細胞(特に樹状細胞からリンパ球を活性化させる系)の活性化にその意義があり、リンパ球レセプターの多様性とは別個に識別応答の系を発達させる必要があった。生物はヒトに到る過程で多くの自然免疫遺伝子を失って環境適応を果たした、と結論づけられる。
