Research Abstract

細胞の形態変化と機能を理解する上で,主要な構成要素であるストレスファイバの力学環境に対する応答性,すなわち力学特性を把握しておくことは大変重要である.最終年度である本年度は,前年度に開発・確立した装置およびストレスファイバ単離手法を用いて以下の2つの研究を行った.
1.ストレスファイバの粘弾性特性の計測
前年度確立した単離方法に従って平滑筋細胞から単離したストレスファイバの引張試験を行い,ひずみ速度が引張特性に与える影響を調べた.ひずみ速度を0.01s^<-1>,0.05s^<-1>,0.1s^<-1>と設定しストレスファイバの引張特性計測を行ったところ,0.01s^<-1>と0.05s^<-1>における張力ーひずみ関係はほぼ線形で両者に顕著な差は見られなかった.一方,生理的なひずみ速度と考えられる0.1s^<-1>では上に凸の非線形な関係が得られた.またストレスファイバの変形初期の強度を示す初期スティフネスに関しても0.1s^<-1>では0.01s^<-1>および0.05s^<-1>に比べ大きくなる傾向が得られた.これらの結果から,ストレフファイバの引張特性にはひずみ速度依存性の閾値が存在する可能性が考えられた.
2.ストレスファイバの局所ひずみ計測
量子ドットを用いてストレスファイバを斑点状に可視化し,これをマーカーとして伸展時におけるストレフファイバ内の局所挙動を観察した.その結果,ストレフファイバは一様に伸展しているのではなくその挙動は局所で異なり,部位によって収縮を示す場合があることが明らかになった.
