Research Abstract

我々が開発を行っている超好熱始原菌Thermococcus kodakaraensisの無細胞翻訳系において、既に耐熱性キチナーゼ(ChiAΔ4)をコードするmRNAを用いたin vitro翻訳が可能となっている。本年度はまず本系を利用したタンパク生産量の向上を進めた。具体的には翻訳反応を長時間行うとS30画分が熱失活している様子が観察されたことから、熱ショックタンパクの転写抑制因子の遺伝子破壊株を用いて無細胞抽出液(S30画分)の調製を試みた。その結果、タンパク合成量が約13%増加し、さらに反応至適温度もこれまでの60℃から65℃へと上昇が見られた。タンパク合成反応のタイムコースを調べた結果、タンパク合成量は反応開始後わずか15分で100μg/mLを突破し、合成量の最大値は115.4μg/mLに達した。続いて本系を転写・翻訳反応系へと展開させることを目的として、mRNAの代わりにDNA、T.kodakaraensisのRNA polymerase、TATA-binding protein、Transcription factor Bを添加してタンパク合成反応を試みたが、これまでに合成タンパクの検出はできていない。この原因には様々なことが考えられるが、主に添加するRNA polymeraseの量が少ないことに加え、S30画分中には多量のゲノムDNA断片が混入しているために、目的タンパク遺伝子以外の転写反応が行われていることが原因であると考えている。RNA polymeraseの精製には労力が必要であることから、特定のサブユニットにタグを付けた株を作製し、そこから多量のRNA polymeraseの精製を行うことを考えている。さらに(期間内には達成することができなかったが)本研究のゴールをとして、オペロンなど複数の遺伝子の転写・翻訳反応を細胞モデルとしてのリポソーム内で行い、これを細胞内で実際に行われている旨「ナノ空間での生命活動」のモデル系としてとらえ、そこから生命現象を形作る基本原理の追求を続けていきたいと考えている。
