褐藻類の鞭毛に局在する新規フラビンタンパク質の機能解析

Research Abstract

本研究は、褐藻類の生殖細胞(遊泳細胞)が有する走光性反応や走化性反応について、そのシグナル受容・変換系を鞭毛構成タンパク質の構造と鞭毛運動の調節機構の観点から解明することを目的とした。これまでの研究において、褐藻カヤモノリ遊泳細胞がもつ2本の異型鞭毛のうち遊泳運動の舵取り機能をもつと推測されている後鞭毛に局在する41kDフラビンタンパク質を精製し、その一次構造と補欠分子(FMN)を同定した。本タンパク質の構造は酵母で発見され機能未知のOLD YELLOW ENZYMEとの類似性を示した。もう一方の推進力を発生するといわれている前鞭毛については、鞭毛軸糸に多数付着する小毛(マスチゴネマ)を構成するタンパク質の同定に成功した。見かけの分子量115kDのタンパク質は珪藻の有性生殖誘導時に特異的に発現する遺伝子と相同であることが判明した。さらに免疫電子顕微鏡観察からこのタンパク質は3部構成のマスチゴネマの中心となるシャフトを構成することを明らかにした。また、マスチゴネマのベース部や先端部を構成する類似タンパク質の候補も見出した。
これらの成果を踏まえ、遊泳細胞の鞭毛の動きとその制御機構を解析するため高速度ビデオ画像による解析を行った。その結果、刺激光に応答して後鞭毛の動きが迅速に変化する様子が明らかになり、また前鞭毛もこれまで考えられていたこととは異なり、細胞運動の推進力を発生するばかりでは無く遊泳方向などの調節にも大きく関与する可能性が示唆された。
これらの解析結果は、走光性や走化性も含めた鞭毛運動とその制御の仕組みの全貌を明らかにするための重要な情報を与え、今後の研究の展開に大きな基礎を築いたと言える。
