ブタ精巣由来の幹細胞株の樹立と遺伝子改変技術への応用

Research Abstract

本研究は、ブタの精巣から幹細胞を分離するとともに、得られた細胞を体外で長期的に培養し、幹細胞培養株からの効率的な遺伝子導入技術への応用を目指している。昨年まで得られた成果から、精巣由来の幹細胞(前原精子細胞)の特異的マーカー(レクチンの一種であるDBA)を見出したので、本年1度はこのマーカーを利用して、分離・精製した前原精子細胞の体外培養を試みた。前精原細胞は、成長因子などの非存在下で体外で活発に増殖し、少なくとも1週間の期間DBAを発現する前原精子細胞として体外で維持された。さらに、これらの細胞は、未分化細胞マーカーである、Nanogをタンパク質および遺伝子レベルで発現し、その他の複数の未分化マーカーも発現することを明らかにした。そこで、ヌードマウスの精巣内にこれらの細胞を注入したところ、精巣内での前原細胞の存在部位で細胞の増殖が認められたことから、これらの細胞が幹細胞としての性質を維持しながら、体外で培養が可能であることを示した。今後、成長因子などの添加によって、体外培養条件を整備することによって、家畜における精巣由来の幹細胞株の作出に展望が開けた。また、本研究では、ブタ精巣の低温下での短期的(1〜2日)な保存と、これらの保存精巣からの前原精子細胞の回収と液体窒素下での凍結保存を試み、前原精子細胞は前述と同様の幹細胞としての性質を体外で維持することを明らかにした。精巣由来の幹細胞は、野生動物や絶滅危惧種など貴重な遺伝資源の保護にも、今後有用な技術となりうると思われた。
