Research Abstract

フラビン含有モノオキシゲナーゼ(FMO)は,NADPH依存的に含窒素,硫黄,リン化合物の酸化を触媒する,一原子酸素添加酵素である.FMO3は成人肝において主要な分子種であり,その機能低下は食物由来成分であるトリメチルアミンの代謝が不十分となり,不快な体臭を放つトリメチルアミン尿症(魚臭症候群)の原因となる.当研究室ではトリメチルアミン尿症の疑いのある日本人から見出した,新規FMO3遺伝子変異を報告した.新規変異であるThr201Lys,Arg205CysおよびMet260Valに着目し,それらのハプロタイプ,頻度ならびに機i能解析を行った.新規FMO3遺伝子変異であるThr201LysおよびMet260Va1はそれぞれ,欧米およびアジアで共通に頻度が高い変異であるGlu158Lys-Glu308GlyおよびVa1257Met変異と同じアリルに存在し,頻度は1%以下であった.Arg205Cysの頻度は約4%であった.大腸菌膜に発現させた,Thr201LysおよびMet260Va1型FMO3のトリメチルアミンおよびベンジダミンN-酸化酵素活性またはメチルパラトリルスルフィドおよびスリンダクスルフィドS-酸化酵素活性におけるVmax/Kmは野生型の約10分の1であった.Arg205Cys型FMO3のN-酸化酵素活性およびS-酸化酵素活性におけるVmax/Kmは野生型の約3分の2および2分の1であった.以上の結果から,新規FMO3遺伝子変異は頻度は低いが,食物由来成分であるトリメチルアミンのみならず,医薬品など各種基質に対する酸化酵素活性を著しく低下させる原因となりうることが明らかとなった.
