Research Abstract

近年、固形がんは単一のがん細胞集団により形成されているのではなく、多剤耐性能を有するがん幹細胞とそれより分化し過剰増殖するいわゆるがん細胞によって形成されていることが解明されてきた。このことは、がん治療におけるがん退縮後の再発機構にがん幹細胞の貢献性が高いことを示唆し、がん幹細胞をターゲットとしたがん治療法の開発の重要性が考慮される。しかし現在、がん細胞がいかなる分子機序によりがん幹細胞化するのかについては全く不明のままである。そこで、成体内の組織細胞と幹細胞の細胞融合のメカニズムに着目し、がん細胞と、造血幹細胞との細胞融合によるがん細胞のがん幹細胞化の可能性につき検討した。昨年度、試験管内では造血幹細胞とメラノーマ細胞株B16の混合培養により、血液細胞と融合したB16細胞が、10-10^3個で、マウスに腫瘍を形成させることから、この融合がん細胞の悪性化を示したが、本年度は生体内で同様の現象が生じるか否かを検討した。CAT遺伝子をLoxPでサンドイッチにしたコンストラクトの下流にGFPを連結した遺伝子をCAGプロモーターの制御下で発現する発現プラスミドを作成しB16細胞に遺伝子導入した。次にアクチンプロモーター制御下でCreリコンビネースを発現するマウス由来の骨髄を用いて、野生型マウスの骨髄を再構築させたマウスを作製した。このマウスに先のCAT-Floxed-GFPを発現させたB16細胞を移植したところ、一部のB16細胞にGFPを発現する細胞が出現した。このGFP陽性B16細胞を回収し、再度10個の細胞をマウスに移植すると、GFP陽性と陰性のB16細胞による腫瘍形成が誘導された。このことは骨髄細胞と腫瘍細胞が融合した細胞は、非常に僅少の細胞でがんを再構築できることを意味し、これらががん幹細胞としての形質を獲得したことが示唆された。
