Research Abstract

(1)IL10ノックアウトマウスをSPFで飼育し、生後4,8週目にH.pyloriを感染させた。その結果、H.pylori感染マウスでは組織学的な腸炎の発症が抑制され、かつマウスの体重減少は抑制された。
(2)一方、生後4週目のマウスにH.pyloriを感染させた。そしてその2週間後からDSSあるいはTNBSを投与して腸炎の作成をこころみた。その結果、H.pylori感染マウスではいずれのマウスにおいても腸炎の発症は抑制された。H.pyloriによる腸炎の抑制作用はDSS腸炎において特に顕著であった。
(3)同様の実験をH.felis感染モデルについてもおこなったが、その腸炎抑制作用は、H.pyloriよりも明らかに強かった。その理由として、H.pyloriがマウス〜早期に排除されるのに対して、H.felisは長く胃内にとどまっていることが考えられた。
(4)CD32とCD80を強制発現させたL細胞と抗CD3抗体の存在下に、腸粘膜から単離したリンパ球を培養して、これらリンパ球からのIFNγ、IL4の産生を見たところ、H.pyloriあるいはH.felis感染マウスからの腸粘膜Tリンパ球は、DSS腸炎、TNBS腸炎いずれの場合においても、そのIFNγ産生が増強しており、逆にIL4の産生は抑制されていた。
(5)以上より、H.pyloriあるいはH.felis感染はIBDの発症に抑制的に作用している可能性があるが、腸粘膜T細胞のTh1シフトを増強しているにもかかわらず、TNBS,DSS腸炎いずれにも抑制をかけることから、その作用はTh1/Th2のシフトといった単純なものではないものと考えられた。
