Research Abstract

近年我々がクローニングしたcoactivator activator(CoAA)は転写活性化因子として機能するほか、RNA結合ドメインであるRNA recognition motif(RRM)を持ち、プロモーター依存性選択的スプライシングを活性化する因子として機能する。Steroid receptor RNA activator(SRA)のようにRNA自体がコアクチベ一夕ーとして機能するものが存在する。RRMを持つCoAA及び、そのスプライシング異性体であり、転写抑制因子であるcoactivator modulator(CoAM)の解析のため、我々は、以下のような新たなin vivo及びin vitroタンパク-RNA結合実験を樹立した。
1)in vivo免疫沈降タンパク-RNA結合アッセイ、
2)磁力ビーズ結合ポリA-RNAを用いた、迅速タンパク-RNA結合アッセイ、
3)発光タンパクであるルシフェラーゼによるmammalian three-hybrid assay、
4)発光標識RNAによるin vitroタンパク-RNA結合アッセイ(Liquid Fluorescent RNA pull down assay)、
5)in vitro GSTタンパク-RNA pull-downアッセイ。
以上のアッセイ及び、既存の実験方法から次の新たな知見が得られた。
(1)CoAAはin vivo及び、in vitroにおいて非特異的にRNAと結合する。(2)In vivo及び、in vitroでCoAAはRNA型コアクチベーターであるSRAと結合するとともに、転写を相乗的に活性化する。(3)CoAMはCBPにより活性化された転写を基底レベルまで抑制するが、このとき、histone acetyltransferase (HAT)活性も抑制される。(4)CoAAによる転写活性化及び、CoAMによる転写抑制はRRMの第1RRMを削除すると消失する。
