Research Abstract

前年度までの研究で、新規生体素材「ゼラチンーGMSM」の埋入により、血管内皮、アストロサイト、ミクロダリアが素材内に新生していることが確認された。その一方、神経細胞の新生は認められなかった。このことは、現方法のままでは神経細胞を有する正常脳の組織再生が不可能であることを示している。この点については、(1)生体素材に間題があった、(2)添加した成長因子(bFGFおよびEGF)が不適切であった、等の可能性が考えられる。
そこでわれわれは、より親水性であり、神経細胞の生着に適していると思われる別の新規生体素材「polydimethylsiloxane-tetraethoxysilane(PDMS-TEOS)」を用いてみた。さらに、神経新生に重要な役割を果たしている血管内皮増殖因子(vascular endothelial growth factor、以下VEGF)を添加する成長因子として用いてみた。具体的には、ゼラチン-GPSMと同様に、ラット脳皮質に欠損を作成し、同部位にあらかじめVEGF(50ng/il)またはvehicleに浸しておいたPDMS-TEOSを埋入、30日後に脳を摘出して組織学的に検索した。
その結果、血管内皮、アストロサイト、ミクロダリアの新生は認めた。一部はBrdU取り込みを認め、物理的に脳の変形を生じて侵入した組織ではないことが分かった。その一方、やはり本方法でも神経細胞の新生は認められなかった。神経新生を生じさせるためには、さらにより最適な成長因子cocktailの作成などを開発していく必要がある。
