Research Abstract

モノリス骨格の表層に各種機能性分子を担持させ、ポリマーモノリスの高機能化を図るとともに、モノリスカラムの特性を最大限に引き出せるシステム・装置の開発を進めた。以下に本年度の具体的な研究成果をまとめる。
1)有機ポリマーモノリスを用いたタンパク質解析用オンライン酵素消化・分離計測システムの開発
プロテオーム解析において高速化のボトルネックとなっているタンパク質の酵素消化に関して、モノリス担体に消化酵素を固定したマイクロリアクターモノリスを試作し、その性能を評価した。数マイクロメートルの微小流路を反応場として用いるモノリスカラム法では、バッチ式反応容器と比べて、反応効率が格段に改善され、従来比1/100の反応時間で定量的にタンパク質を消化できた。本カラム法では、10分以内に酵素消化を達成できることから、酵素消化と分離分析、さらに質量分析の各工程をオンラインで接続でき、"Just in time"でタンパク質の解析を行える。
2)マイクロ固相抽出・濃縮デバイスの開発
前処理の高速化、簡便化を目指して、モノリス型マイクロ固相抽出・濃縮デバイスの開発を検討した。除粒子、清澄ろ過等の前処理に汎用されている市販のシリンジフィルターを容器として用い、その内部スペースにメタクリル酸グリシジルベースのモノリス担体を調製した。その表層にエポキシ基の開環反応を利用して8-キノリノールやイミノジ酢酸を固定することにより、微量金属元素捕集用のキレート交換モノリスを作製した。本デバイスのキレート交換容量は1-2μmol/tipであり、200mLの海水に含まれる銅や亜鉛、さらに超微少量の希土類元素を定量的に捕集・回収することができた(100倍濃縮)。その結果、ICP-MSにより、これまで測定が困難であったsub-pptレベルの超微量元素を精度よく定量することに成功した。
