Research Abstract

植物の生育に必須な元素の一つであるリンは、資源枯渇の問題に直面しつつあるとともに、環境への負荷の問題も抱えている。土壌中に蓄積する有機態リンのうち最も高い割合を占めるフィチン酸を有効利用することにより、これらの問題の解決につながると考えられる。本研究は、ルーピンが示すフィチン酸利用能を解析し、これを応用することを目指して実施した。
1)分泌性酸性ホスファターゼ遺伝子LASAP2のフィチン酸分解能の利用
ルーピン根由来分泌性酸性ホスファターゼをコードするLASAP2によるフィチン酸利用能を有効活用する目的で、低リン条件特異的誘導型プロモーターの探索を行った。LASAP2プロモーターのdeletionシリーズの構築に成功した。また、イネで低リン条件に強く応答するOsP11遺伝子のオーソログがルーピンで少なくとも4種類存在しており、プロモーター構築の良い候補となることが示された。
2)フィターゼ活性を有する新規酸性ホスファターゼホモログLASAP3の機能の利用
昨年度までに分泌性の可能性が示唆されたLASAP3を、CaMV35Sプロモーター制御下でタバコに形質転換を行った。その結果、植物由来分泌型フィターゼを高発現する植物の育成に成功した。さらに、フィターゼ活性を有する可能性のある酸性ホスファターゼホモログcDNAの単離に新たに成功し.LASAP4と命名した。
(3)ルーピン根圏から単離したフィチン酸資化性細菌の利用
ルーピン根圏由来のフィチン酸資化性細菌を多く含むBurkholderia spの単離菌株の植物との相互作用を調査するため、共培養系を構築した。共培養した結果、単離菌株のうちの一つは植物根の側根の伸長を促進するが、別の株は強く主根の伸長を阻害する効果を示した。これらの植物-微生物間相互作用は、水溶性物質を介して行われていることを示した。
