酵母を用いたステロールのメタボロミクスと輸送の分子機構の解明とその応用

Research Abstract

1. 出芽酵母Saccharomyces cerevisiaeより調製した形質膜画分をドナー膜とし、小胞体画分をアクセプター膜としたin vitroのオルガネラ間ステロール輸送の再構成系を構築して、その機構について解析を行った。出芽酵母のオキシステロール結合タンパク質の1つであるOsh4pが形質膜から小胞体へのステロール輸送に関わることが強く示唆された。
2. 細菌由来のステロールアシルトランスフェラーゼを酵母内で発現させ液胞に局在させることにより、液胞へのステロール輸送をエステル化によって評価するステロール輸送解析系の構築を進めた。本アシルトランスフェラーゼを液胞に局在させた酵母から単離した液胞膜と、放射性同位体標識したエルゴステロールを含むP12膜画分を用い、in vitroで液胞へのステロール輸送を解析する系を構築した。
3. 酵母のオキシステロール結合タンパク質の機能や役割分担を明らかにするため、すべてのOSH遺伝子が破壊された遺伝的背景で、Osh2〜7pとGFPとの融合タンパク質を単独で高発現する株を作製して、Oshタンパク質の局在や細胞内のステロール分布の観察を行った。この系により、個々のOshタンパク質のステロール輸送における役割についてin vivoおよびin vitroで定量的に解析することが可能となった。
4. 脂質合成や含量を培地の成分により制御できる株を用いて各種リン脂質のESI-MS/MSを用いたメタボローム解析を行い、培養条件によりリン脂質プロファイルが変動することを示した。
