泌乳前期の乳牛におけるエネルギーバランスの改善と泌乳量に対するグレリンの生理作用

Research Abstract

本研究では、反芻家畜の泌乳とエネルギーバランスの制御におけるグレリンの作用を明らかにすることを目的とする。平成16年度において、泌乳前期のザーネン種山羊にヤギグレリンを連続投与すると泌乳量が増加することを明らかにした。今年度は、その際の摂食とエネルギー出納、インスリン感受性について解析した。摂食量はグレリン注入により増加した。呼吸試験と消化試験の結果から得られたエネルギーバランスは、対照区、グレリン区ともネガティブであったが、グレリン区では改善する傾向にあった。グレリン注入によって生体のインスリン感受性は低下した。次に、泌乳最盛期のホルスタイン種経産泌乳牛4頭を用いて、グレリンが乳生産、摂食およびエネルギー出納に及ぼす影響を調べた。本実験では、頸静脈カテーテルからウシグレリンの連続注入を行うとともに、消化試験と呼吸試験を行った。グレリン注入は、泌乳最盛期の乳牛の摂食と泌乳量に影響しなかった。飼料中脂質成分の消化率がグレリン注入によって低下したため、飼料エネルギー(GE)の消化率もわずかではあるが低下した。エネルギーのRetentionは、グレリン注入によって低下する傾向にあったが、本実験では泌乳牛のエネルギーバランスはグレリン投与の有無に関わらずポジティブであった。また、泌乳最盛期の乳牛に対するグレリンの投与は、生体のインスリン感受性を抑制した。本研究では、グレリン注入によって泌乳前期の山羊の摂食量と泌乳量は増加し、同時にエネルギーバランスも改善したが、泌乳最盛期の乳牛では摂食と乳生産には変化が認められなかった。動物種や条件に違いがある実験結果の比較から結論を導き出すことには慎重であるべきだが、少なくとも泌乳期や栄養状態によってグレリンの乳生産やエネルギー出納に対する生理作用は変化する可能性が示されたと考えられる。
