Research Abstract

本年度は、新たな骨格転移反応を用いた二量体型アルカロイド、ハプロファイチンの左部4環性骨格のモデル化合物の構築に成功した。また、ハプロフィチンの右部を構成しているアスピドファイチンに関して、Fisherインドール合成法を用いる全合成を達成することができた。前者については、まず、文献既知のテトラヒドロ-β-カルボリン誘導体から1,2-ジアミノエテン構造を有する基質を合成した。この化合物をmCPBAで処理したところ、二重結合のエポキシ化と続く窒素原子からの電子供与による位置選択的なエポキシドの開環、ピナコール転移と類似の骨格転移反応が進行し、アミナールと架橋ケトン部を有するハプロファイチンの左部4環性骨格の構築に成功した。位置選択的なエポキシドの開環には窒素の保護基の選択が重要であり、ニトロベンゼンスルホニル基では骨格異性体の混合物が生成したのに対し、Cbz基を有する基質は選択的に望みの生成物を与えた。なお、生成物の構造は、Cbz基を脱保護した後、N-メチル化を行って得た化合物のX線結晶構造解析を行うことにより決定した。また、アスピドフィチンの合成について、ダンジェロらの不斉共役付加反応を用いて合成した光学活性3環性ケトンのFischerインドール合成法を鍵工程とする全合成を達成することができた。以上の検討により、上記、骨格転移反応を用いて合成したハプロファイチンのモデル化合物の誘導体に対してFisherインドール合成を適用することで右部アスピドファイチン部の構築を行い、ハプロファイチンの合成を行う合成戦略の実現性を示すことができた。
