Research Abstract

癌抑制遺伝子p53は細胞周期の制御や、アポトーシス誘導に重要な役割を果していることが明らかになってきたが、その生理機能と調節機構の詳細ははっきりしない。特に、p53転写活性の調節についての情報は極めて少ないのが実状である。我々は、リン酸化酵素プロテインキナーゼCファミリーの一員であるPKCdeltaが翻訳後修飾だけでなく、転写レベルでもp53の発現を制御していることを見いだした。p53のプロモーターを用いたレポーターアッセイの解析から、PKCdeltaがp53のプロモーター領域のうちCPEと呼ばれる配列を介して、p53の転写活性を促進することを明らかにした。さらに、我々はCPEに結合している転写因子はアポトーシス促進因子Btfであることを見いだした。DNA損傷によりPKCdeltaはBtfと複合体を形成しており、PKCdeltaによって活性化されたBtfはCPEと結合し、p53のプロモーター活性を誘導するが、PKCdeltaの阻害剤によりBtfとCPEの結合が顕著に抑制された。その結果、p53のmRNAと蛋白質レベルの発現も明らかに減少した。また、PKCdeltaの阻害剤やBtfのsiRNAを用いることで、DNA損傷によるp53依存性アポトーシス誘導が著しく低下した。これらの結果から、PKCdeltaはBtf依存性p53転写制御を介して、DNA損傷によるアポトーシスを誘導することが明らかになった。
