生細胞を用いたリアルタイムイメージングによるヘルペスウイルス制御因子の機能解析

Research Abstract

ダイナミックな動態を示すウイルス因子の解析には、同一の生細胞をリアルタイムで、連続的に観察するといったウイルス因子の動態解析が極めて重要となってくる。本研究では、最新のイメージング技術を駆使して、生きた感染細胞において、HSV(herpes simplex virus)がコードする様々なウイルス因子の動態を可視化する。そして、従来の研究手法では見いだせずにいた、ウイルス因子の新たな側面を明らかにし、HSV増殖機構の新たなパラダイムを構築することを目的とする。本年度は、HSVのカプシドタンパク質UL35、局在が異なる2つのテグメントタンパク質UL49、エンベロープタンパク質gBをそれぞれVenus、mPFR、ECFPで標識した組み換えウイルスYK608を用いて、感染細胞の詳細な時空間的リアルタイムイメージング解析を行った。その結果、細胞の細胞の基底面にウイルス粒子が成熟する場が複数形成されることを明らかにした。また、その場には宿主細胞のオルガネラであるトランスゴルジネットワークのマーカーが特異的に集積することが明らかになった。つまり、HSVの最終エンベロープ獲得の場がトランスゴルジネットワークであることが明らかになった。
