Research Abstract

平成19年度は(1)Foxp3の発現解析、(2)Foxp3^+制御性T細胞の機能解析、(3)Foxp3による制御性T細胞分化・機能の制御メカニズム、につき研究を進めた。(1)および(2)に関しては、Foxp3^+T細胞を生細胞レベルで解析し、これを特異的に除去するためのマウスモデル(Foxp3-ires-hCD2-CD52ノックインマウス)確立し、抗hCD2抗体によりFoxp3^+T細胞を単一生細胞レベルで同定・分離できること、抗hCD52抗体投与によりこれを特異的に除去できることを示した。さらに、Foxp3は制御性T細胞のみならず通常のナイーブT細胞にも低レベルで発現すること、そして制御性T細胞におけるFoxp3発現は必ずしも安定ではなくdown-regulationされうるものであることを明らかにし、Foxp3は必ずしも制御性T細胞の特異的マーカーではない可能性を示唆した。(3)に関しては、引き続きヒト自己免疫疾患IPEX患者において見つかっているFoxp3遺伝子変異の制御性T細胞の発生・分化と機能に与える影響の解析を進め、1アミノ酸置換を引き起こす3つの代表的点変異に関してノックインマウスを作製し、解析を進めた。その結果、これら変異マウスはFoxp3欠損マウスと同様の自己免疫疾患を発症し、IPEXの原因がこれら変異にあることを証明した。さらに、これらのうち1つの変異ノックインマウスにおいては、変異体Foxp3発現T細胞は正常に発生し、そして細胞表面マーカーの発現様式など野生型と同様の性質を示した。このことは、この変異マウスが自己免疫疾患を発症する原因は制御性T細胞の分化異常というよりも機能異常である可能性を示唆しており、Foxp3により制御される標的遺伝子のうちこの変異により発現が影響を受ける遺伝子を同定することで制御性T細胞の機能を担う遺伝子を同定できる可能性が考えられた。
