Research Abstract

p62(A170)過剰発現PC12細胞,p62(A170)欠失皮膚線維芽細胞を用いた細胞レベルでの研究
異常Httを培養細胞に導入する.とp62(A170)発現量を増加させることを明らかにし,Httの細胞内処理にp62(A170)が要求されることが示唆された.p62-DsRed過剰発現PC12細胞を構築し、細胞内での挙動を観察した.タンパク分解抑制およびHttやsynucleinなど異常タンパク発現時に凝集体を作りやすい性質が認められた.
この結果をもとに,p62(A170)欠失皮膚線維芽細胞および正常細胞に,GFP付加異常Htt,GFP付加正常Httを発現させ,凝集体の形成過程の検討,p62(A170),ユビキチンの免疫染色等を行った.
患者由来細胞ではプロテアソーム活性が低下していること,カテプシン活性が上昇していることを確認した.正常者5名,ハンチントン病(HD)患者8名の皮膚線維芽細胞を用いて,p62(A170)および細胞死おタびダンパク分解に関与する分子の発現量を検討し,発現量に差があることを確認した(投稿準備中).また,遺伝子導入細胞と同様の細胞内構造物が患者由来の線維芽細胞でも認められた.
p62(A170)KOマウスとHDモデルマウスの交配による,臓器レベル,個体レベルでの研究.
上記2種のマウスの交配により,p62(A170)を欠失し,かつ異常Httを有するモデルマウスが生誕する.現在,2種のマウスの交配に成功し,目的とするマウスがすでに生誕,形態学(電顕を含む),免疫組織化学による検討を行っている.個体レベルの現象として歩行異常などの神経症状,寿命の長短も検討中であるが,少数個体での検討では,優位な差を認めていない.さらに別系統のHDマウスとの交配実験を他施設と協同で進行中である.
