Research Abstract

本研究では小脳バーグマングリアに発現するAMPA受容体の詳細な細胞内局在を明らかにし、神経細胞でのAMPA受容体局在制御機構との比較を行い、その細胞表面発現機構へのstargazinファミリー蛋白の関与を明らかにすることを目的として研究を行った。
まず、小脳バーグマングリアに発現するAMPA受容体のサブユニットであるGluRα1とGluRα4の局在解析を行った。その結果、AMPA受容体は平行線維とプルキンエ細胞の興奮性シナプスの周囲に豊富に存在し、特にシナプス間隙近傍のバーグマングリア細胞膜に局在していた。このことは放出されたグルタミン酸をバーグマングリアが効率よく受容するための局在制御機構が働いていると考えられる。さらに、stargazinファミリー蛋白mRNAの発現解析の結果、小脳バーグマングリアではg4とg5が発現していることが明らかとなった(Fukaya et al.,2005 Neurosci Res)。さらに抗体を用いて局在解析を行ったところ、g5はAMPA受容体と同様に平行線維-プルキンエ細胞シナプス間隙近傍のバーグマングリア細胞膜に局在していた。このことはg5がAMPA受容体の局在を制御する可能性を示唆している。また、g8の局在解析およびノックアウトマウスの解析の結果、g8はAMPA受容体が豊富なシナプス後膜肥厚部に多く局在し、棘突起や樹上突起の細胞膜にも存在していた。g8ノックアウトマウスではシナプス部および棘突起や樹上突起の細胞膜でのAMPA受容体が減少していた。このことからg8はAMPA受容体の局在制御に深く関与していることが明らかとなった(Fukaya et al.,2006,Eur J Neurosci)。
以上の結果から、バーグマングリアでのAMPA受容体の局在制御には神経細胞と同様にstargazinファミリー蛋白が関与する可能性が示唆された。
