Research Abstract

本年度は主に体細胞クローン技術により作製された初期胚を用いて胎盤関連遺伝子の定量RT-PCRによる発現量の解析を行った。正確な発現データを取るために、解析には単一胚を検体として用いることとした。単一胚の転写産物は非常に微量であるため、IVTによる増幅方法、SPIAプライマーを用いるRibo-SPIA増幅方法の検討を行った。結果としてIVTによる増幅法が増幅をしないサンプルと比較して定量解析の再現性があることや、多量の増幅産物が得られることから、遺伝子発現量の解析にはIVT増幅サンプルを用いた。定量に利用した遺伝子は未分化栄養膜細胞の増殖因子であるFGF4、その他の胎盤形成に関わる遺伝子であるFGFR2(IIIb、IIIc)、細胞の多能性維持に関与する転写因子であるOct4、同様に未分化細胞での発現が確認されている転写因子Sox2そして内部コントロールとしてChuk、Gapdh、Hprtの測定値をを利用した。その結果、調べた全ての検体(体外受精胚9個、卵丘細胞クローン胚9個)について、FGF4の発現が確認され、また発現量もGapdhをコントロールとした場合を除き、体外受精胚に比べ高いという予想外の結果が得られた。また他の遺伝子FGFR2(IIIb,IIIc)、Oct4、Sox2もクローン胚の方が有意に高いことが明らかとなった。Gapdhについては、他の二種類のコントロールに対しクローン胚における絶対値が2倍以上高く、このことが発現量の差を打ち消してしまうためと考えられた。すなわち、FGF4の発現低下が胎盤形成不全の原因になっていると予想したのとは全く逆で、総じて遺伝子発現量は体外受精胚よりクローン胚の方が高いことが明らかになった。現在は、この遺伝子発現亢進がどのように今後の着床、胎盤形成に関わって来るのかを明らかにすべく、抗体染色を用いた組織的解析を行っているところである。
