Research Abstract

動脈硬化初期過程における我々の研究から,酸化LDLは血管内皮細胞間隙分子の発現制御(PECAM-1の増強,VE-cadherinの抑制)により細胞間隙を単球が浸潤し易い状態に変化させ,それにより,単球の接着→浸潤開始のstepが特異的に促進されることが明らかとなった(Hashimoto K et al., Atherosclerosis 2007,in press).本研究では,単球浸潤時のPECAM-1,VE-cadherinの分子動態を生細胞において直接観察することを目的として,当初は3種類の分子(酸化LDL,単球CD14,VE-cadherinor PECAM-1)の同時抗体染色を想定していた.しかし,膜貫通タンパクであるPECAM-1(VE-cadherin)の抗体による細胞外ドメイン標識は,単球浸潤の障害になる可能性がある為,代替法として,これらの分子の細胞内ドメインに別色の蛍光タンパク(GFP, DsRed)を付加した組み換えベクターを作製し,内皮細胞及び単球に発現させることにより,細胞外での分子相互作用を阻害せずに両分子の動態を直接観察することを試みた.本年度は,PECAM-1に着目し,常法(RNA抽出,逆転写,PCR増幅,ベクターligation,コロニー選択,プラスミド精製,塩基配列確認)によりPECAM-1-GFP(緑)及びPECAM-1-DsRed(赤)の発現ベクターを構築した.次に,様々な遺伝子導入法を検討した結果,Nucleofection法(Amaxa社)により高効率・低毒性にてベクターを導入・発現させることに成功した.現在,これらを発現させた細胞の機能確認(形態,増殖能,発現分子の局在確認,分子量確認等)を行っており,これらの細胞を用いて,共焦点レーザー顕微鏡による3次元浸潤動態観察を行い,両分子の役割を直接的に検証する予定である.
