不斉自己増殖するキラルデンドリマーの開発

Research Abstract

分子内に不斉自己増殖するピリミジルアルカノール部位を複数有する化合物の効率的合成法の検討を行った。ピリミジン環の導入に際し、従来法ではシリルクロリドヘアルキニルグリニャール試薬を作用していた。今回、カルボシラン骨格の末端をシリルアセチレンとし,2位にクロロ基を有するピリミジン部位と薗頭カップリング反応を行うことにより高効率的に合成することを可能とした。
本法を用いて分子内に高い不斉自己触媒活性部位を4つ有する(S4)-テトラキス(ピリミジン)テトラアルカノールを合成した。ラセミ体と光学活性体を混合することにより異性体比(S_4)/(S_3,R)/(S_2,R_2)/(S,R_3)/(R_4)=6/25/38/25/6、鏡像体過剰率0.5%eeの化合物を不斉自己触媒として用い、4つの不斉中心を1回の反応で構築可能な不斉自己触媒反応を行った。対応するテトラカルバルデヒドに対し5-20mol%の不斉自己触媒を用いジエチルエーテル中,0℃で8.8等量のジイソプロピル亜鉛を作用した。1回の反応で異性体比は(S_4)/(S_3,R)/(S_2,R_2)/(S,R_3)/(R_4)=28/26/21/16/9、鏡像体過剰率は56%eeに向上したテトラアルカノールが得られた。さらに、得られたテトラアルカノールを次の反応の不斉自已触媒として用い,反応を行った結果、(S_4)体が著しく増殖し、5回目の反応では異性体比(S_4)/(S_3,R)/(S_2,R_2)/(S,R_3)/(R_4)=98/1/1/0/0で91%の収率で生成物が得られた。以上テトラキスピリミジルアルカノルを用いる不斉自己触媒反応において4つの不斉中心を1回の反応で構築することが可能な不斉自己触媒反応を見出した。さらに低鏡像体過剰率の触媒を用い連続的に反応を行うことにより,顕著な不斉の向上、さらには異性体比の改善を伴う不斉自己触媒反応を実現することに成功した。
