全反射赤外分光法による生体適合性高分子へのタンパク質吸着のその場追跡

Research Abstract

高分子フィルムにタンパク質水溶液を接触させ,固液界面の全反射赤外スペクトルを時間分解測定することで,生体適合性高分子フィルムヘタンパク質が吸着する過程をその場追跡した.これにより,被吸着高分子の違いによる吸着過程と吸着構造との相関を議論した.また,乾燥フィルムが含水する過程を時間分解全反射赤外測定することで,タンパク質吸着に影響を与えるといわれている高分子フィルムの水和構造を議論した.
抗血栓性高分子であるpoly(2-methoxyethyl acrylate)(PMEA)は,これまでの研究により,不凍水,中間水,自由水と呼ばれる三種類の水和水を持っことが知られている.本研究によりPMEAへの含水過程を時間分解全反射赤外測定したところ,三種類の水に帰属されるバンドが観察された.
このようにして得られた時間分解スペクトルを解析して有用な情報を抽出するために,多変量解析を応用した摂動相関二次元相関分光(PCMW2D)法を開発した.また,測定スペクトルから予測される水和構造の妥当性を確認するために,量子化学計算によるシミュレーションスペクトルを計算し,測定スペクトルとの比較を行った.
これらの解析結果から,PMEAの不凍水は,側鎖カルボニル基と水素結合しており,この強い束縛が不凍水を-100℃まで結晶化させないことがわかった.また,中間水は,PMEAの化学構造に特徴的な側鎖末端メトキシ基に,数個の水分子がクラスターを形成して水和していることがわかり,この特徴的な水和構造がタンパク質の吸着を抑制するなど,血液適合性に重要な役割を果たしていることがわかった.さらに,自由水はバルク水と類似した水素結合ネットワーク構造を持つことがわかった.
