Research Abstract

我々のグループは、鳥類のウズラの脳から生殖腺刺激ホルモンの放出を抑制するペプチドを単離・同定し、生殖腺刺激ホルモン放出抑制ホルモン(Gonadotropin-inhibitory hormone ; GnIH)と名付けた(BBRC 275:661-667,2000)。さらに、最近このGnIHレセプターを分子生物学的手法により同定し、その結合特異性を生化学的手法により明らかにした(J.Endocrinol.184:257-266,2005)。
本研究の目的は、新規視床下部ホルモンであるGnIHの作用機序を明らかにすることである。そのためには、GnIHレセプターが発現している細胞を同定する研究が必要不可欠である。
そこで、GnIHレセプターmRNAの発現部位をin situハイブリダイゼーション法により詳細に解析した。さらに、GnIHレセプターの発現部位と脳下垂体ホルモン産生細胞や生殖腺刺激ホルモン放出促進ホルモン(Gonadotrobin-releasing hormone ; GnRH)産生細胞の関係を解析するために、GnIHレセプターのin situハイブリダイゼーションと脳下垂体ホルモンやGnRHの免疫組織化学法を同一切片上で行った。
その結果、GnIHレセプターのmRNAは、脳内の広い領域に発現していることが明らかとなった。特に高発現を示した部位は、GnRHニューロンを含む視索前野や視床下部領域、さらには脳下垂体であった。特に脳下垂体では生殖腺刺激ホルモンが発現している細胞にGnIHレセプターが共発現していた。この結果は、GnIHは脳下垂体細胞に直接作用し、生殖腺刺激ホルモヒの放出を抑制することを示している。加えて、GnIHは視索前野や視床下部を介して間接的に生殖腺刺激ホルモンの放出を調節している可能性も示された。
尚、GnIHレセプターに対する抗体の作成を試みたが特異的な抗体を得るに至らなかった。
