Research Abstract

真核生物のグループの一つであるイクチオスポラに属するPseudoperkinsus tapetisと命名された原生生物は,スペイン北西部ガリシア地方の食用アサリから初めて分離された。本研究において,鹿児島県野間岬沖の水深約200mから回収された鯨骨に付着した2枚貝ヒラノマクラのエラ組織より真核微生物を分離し,核コードのSSU rRNA遺伝子の塩基配列情報に基づいて,それをP.tapetisとして同定した。スペイン産および本研究によって発見された日本産のP.tapetisのSSU rRNA遺伝子(1751b.p.)の配列を比較したところ,1塩基の置換と1塩基のindelが見いだされたのみで,ほとんど違いはなかった。一方,SSU rRNA遺伝子より進化速度の速いITS-rDNAを比較したところ,ITS1領域で10塩基,ITS2領域で8塩基の置換が認められ,スペイン株と日本株は集団レベルで遺伝的に異なっていることが示された。今回の結果から,P.tapetisは世界中に存在し,様々な2枚貝に寄生することが可能であることが示唆された。さらに、ITS-rDNA領域の塩基配列情報を用いて,宿主からP.tapetis細胞を特異的に検出するPCR法も確立した。この特異的PCR検出法によって、水槽中で飼育されたヒラノマクラにもP.tapetisが存在することが明らかとなった。上記の通り,P.tapetisは2枚貝の寄生虫である可能性が高く,その生態学的な知見を得ることは重要である。本研究において確立された検出方法はP.tapetisの生態学的研究における強力なツールとして利用出来るものと考える。
