Research Abstract

セラミド(Cer)は近年、細胞内脂質セカンドメッセンジャーとして細胞周期の停止やアポトーシスの誘導に関与する事が明らかになり注目を集めてきた。しかし、Cerの細胞内における直接のターゲットが明らかになっていない為に、Cerの代謝産物や代謝酵素のCerシグナリングにおける役割も最重視されつつある。我々はCer代謝酵素であるセラミドキナーゼ(CERK)に注目し、本酵素の生理的役割を明らかにすべく本研究を遂行している。本年度我々はCERK遺伝子を欠損したマウスの解析をすすめ、その中で、CERKがマウス小脳プルキンエ細胞に高く発現している事を見いだした。さらに、CERK遺伝子の欠損により、通常の運動能力等には支障が無いが、情動性に異常が見られる事を明らかにし報告した(Biochem. Biophys. Res. Commun., 2007)。また、前年に開発したCERK阻害剤K1の酵素阻害様式を明らかにした(Biosci. Biotech. Biochem., 2007)一方、神経細胞以外でのCERKの解析もすすみ、CERKが前骨髄性白血病細胞HL60の分化時に発現が変動する事を見いだし報告した(In vitro Cell. Dev. Biol.-Anim. 2007)。
さらに、CERKをCerの膜上での動態を解析するツールとして使用し、生きた細胞膜上でCerが自由に脂質二重膜を通過する事をはじめて明らかにした(Biochem. Biophys. Res. Commun., 2007)。
本年度は、本研究計画の最終年であったので、これらの内容や過去のCERKに関する報告を総説として発表した(Prog. Lipid Res. 2007)。
