Research Abstract

秋播き性パンコムギは出穂の低温要求性を決める遺伝子座であるVrn-1について全て劣性アリルを持ち、春播き性パンコムギはVrn-1について優性のアリルを持つ。また特に劣性のvrn-A1アリルを持つ系統は優性のVrn-A1アリルを持っ系統よりも冬期の凍結低温耐性に優れているとされる。低温凍結耐性を決める主働遺伝子Fr-1はVrn-1に連鎖して座乗する。この生育環境に対応した2つの遺伝子座(Vrn-1とFr-1)のアリル間に連鎖関係があるのか、あるとしてパンコムギの持つ3つのゲノムA,B,D全てにおいて言えるか、祖先野生種ではどうか、Vrn-1とFr-1がどの程度コムギの適応に重要なのか、これらに答えることが本研究の目的であり、本年度は特にFr-1の機能についての解析を行った。
Fr-1が制御するFr-2座に座乗し転写因子をコードするWCBF2遺伝子について、これを恒常的に発現させた形質転換タバコを用いて凍結耐性発揮への直接的証明を行い下流のCor/Lea遺伝子のいくつかの転写を直接活性化することを明らかにすると共に、WCBF2がDゲノム特異的コピーであり他のCBF転写因子遺伝子と異なり進化的に中立であることを明らかにした。同様にFr-1に制御される転写因子WDREB2やWLIP19についてもそのス下レス耐性への関与と下流のCor/Lea遺伝子発現の活性化を示した。一方でWLIP19に相同性のあるWABI5についてはストレス耐性への関与と下流のCor/Lea遺伝子発現の活性化は示されたが、Fr-1とは独立に機能していると考えられた。この他にもミトコンドリアで活性酸素消去に関与するAOX遺伝子が凍結耐性に関与し品種間差異に貢献していることを示した。このFr-1とABAとの関係については総説にまとめ、Vrn-1の変異についてもコムギの栽培化と品種分化の観点から和文の論文にまとめた。
