Research Abstract

天然および非天然の生物活性物質には複雑な環骨格をする化合物が多いため、複雑な複糸環骨格の効率的な合成法の開発は最重要かつ緊急の研究課題である。本研究は,環境に配慮したタンデム型閉環反応による複雑な生物活性複素環の一挙構築法の開発を目的として、アレン系及びアセチレン系化合物を用いた連続閉環反応を検討し、平成18年度に以下の研究成果を得た。
(1)三成分カップリング-環化カスケード反応によるインドール誘導体の合成
報告者は、エチニルアニリンに対して銅塩存在下、ホルムアルデヒドとアミンを反応させると、三成分カップリング-閉環反応が効率よく進行し、アミノメチルインドール誘導体が一挙に得られることを見出した。アミン成分としてプロモベンジルアミン誘導体を用い、得られたインドールに対してパラジウム触媒を作用させると、縮環型インドールが簡便に合成できることも併せて明らかにした(Angew.Chem.Int.Ed.2007,46,2295)。
(2)プロモアレンをアリルジカチオン等価体として用いた新規連続閉環反応の開発
報告者はこれまでの研究において見出したプロモアレンがアリルジカチオン等価体として機能する新しい反応性を利用し、今回分子内にスルファミドを有するプロモアレンを用いたタンデム型閉環反応を検討した。その結果、生物活性化合物に多く存在する環状スルファミド骨格の一挙構築に成功した(Chem.Eur.J.2007,13,1692)。
(3)試薬・触媒を用いないアレンと多重結合の環化付加反応
触媒や試薬を用いずに進行する環化付加反応は、すべての原子を有効利用するアトムエコノミーに優れた反応である。今回、アレンと多重結合間の環化付加反応を検討し、加熱のみによって縮環型シクロブタン誘導体が高収率で得られることを始めて見出した(J.Org.Chem.2007,in press)。
