塩基による電子移動触媒作用を利用した強力な新規抗酸化物質の開発

Research Abstract

研究代表者は平成17年度までに、抗酸化物質によるラジカル消去反応が、電子移動を経由して進行する場合には、塩基が中間体として生成する抗酸化物質由来のラジカルカチオン種を安定化することにより、ラジカル消去反応を顕著に加速することを見い出した。そこで平成18年度は、天然抗酸化物質よりも強力で毒性の少ない新規抗酸化物質を開発するため、前年度に得られた成果をさらに発展させ、分子内に塩基性部位を有する天然抗酸化物質誘導体を合成した。有機合成の手法により、水溶性ビタミンE類縁体であるトロロックスに種々の長さの炭素鎖をもつピリジン誘導体を導入し、3種類の新規ビタミンE誘導体を得た。次に、嫌気性条件下、メタノール中、25℃で、得られた新規ビタミンE誘導体とDPPH(2,2-diphenyl-1-picrylhydrazyl)ラジカルとの反応をストップトフロー分光測定装置を用いて追跡した。その結果、いずれの新規ビタミンE誘導体も非常に効率良くDPPHラジカルを消去することがわかった。DPPHラジカルに由来する吸収の減少を速度論的に解析することにより、新規ビタミンE誘導体によるDPPHラジカル消去の反応速度定数を決定した。その結果、3種類の新規ビタミンE誘導体のうち2つは、ビタミンEの活性中心モデルよりも2〜3倍のDPPH消去活性を有することがわかった。これは、中間体として生成するビタミンE部位由来のラジカルカチオンに塩基性のピリジン部位が配位することによって安定化し、ラジカル消去反応を加速していると考えられる。以上の結果から、天然抗酸化物質であるビタミンEの活性中心モデルに、電子移動触媒作用を示す塩基性ピリジン部位を導入することにより、天然抗酸化物質よりもラジカル消去活性が高い新規抗酸化物質を開発することに成功した。
