新規生体素材を用いた脳の組織再生の試み

Research Abstract

前年度までの研究で、新規生体素材「ゼラチン-GMSM」の埋入および切片の作成までは終了している。その切片を使用しての各種細胞マーカー(tomato-lectin、NeuN、GFAP、Ibal)での免疫染色により、生体素材内の細胞同定を行った。その結果、血管内皮、アストロサイト、ミクログリアは多数同定され、それらのうちの一部は分裂マーカーも陽性であった。その一方、神経細胞の新生は認められなかった。このことは、現方法のままでは神経細胞を有する正常脳の組織再生は困難であることを示している。
そこでわれわれは、より親水性であり、神経細胞の生着に適していると思われる別の新規生体素材「polydimethylsiloxane-tetraethoxysilane(PDMS-TEOS)」を用いてみた。さらに血管内皮増殖因子(vascular endothelial growth factor、以下VEGF)を浸して、埋入したPDMS-TEOSへの神経新生を促すことを試みた。具体的には、ゼラチン-GPSMと同様に、ラット脳皮質に欠損を作成し、同部位にあらかじめVEGF(50ng/il)またはvehicleに浸しておいたPDMS-TEOSを埋入、30日後に脳を摘出して組織学的に検索した。
その結果、血管内皮、アストロサイト、ミクログリアの新生は認めたが、やはり本方法でも神経細胞の新生は認められなかった。神経新生を生じさせるためには、さらにより最適な成長因子cocktailの作成などを開発していく必要があると思われる。
