Research Abstract

これまで,椎間板の変性変化には椎間板細胞のアポトーシスが関与している可能性があることが示唆されていたが,その詳細なメカニズムは依然不明であった.本研究ではアポトーシスにおいて直接的な役割を果たす遺伝子群と,椎間板細胞を用いた遺伝子導入実験を行い,椎間板細胞のアポトーシス誘導機構の解析とこれらが椎間板変性に及ぼすメカニズムについての検討を行った.
ラットアポトーシス抑制遺伝子:Bcl-2に対する最適化遺伝子合成を行い発現プラスミドを構築した.これをラット椎間板より採取拡大培養した椎間板細胞に遺伝子導入した.遺伝子導入48時間後に血清飢餓を開始しアポトーシスを誘導した.Bcl-2を過剰発現した系では,コントロール群に比べTunne1法にて断片化DNAが減少し,FACSにおいてもアポトーシス陽性細胞の減少が確認された.同時にRT-PCRにてCaspase3の発現量が低下していた.つまり,Bcl-2の過剰発現はCaspase3によるアポトーシスの実行を抑制することが明らかになった.椎間板細胞アポトーシスを遺伝子レベルで抑制することで椎間板変性に対する初期治療に応用できると思われる.
