Outline of Final Research Achievements

To elucidate the molecular mechanismof G13-RhoGF signaling pathway, we tried to determine the structure and Ga13-RhoGEF complex, and dynamic mechanism of Ga13-RhoGEF interaction. As we could not determine the complex strucuture of G13-RhoGEF, we could not peform detailed interraction mechanism of G13 and RHoGEF.

Academic Significance and Societal Importance of the Research Achievements

GPCRシグナル伝達系は、様々な整理機能を調節してることが知られているが、G12／13-RhoGEF伝達系については、未だ十分に分子機構が解明されていない。この伝達系の分子機構を理解するために、そのG12／13-RhoGEFの複合体の構造解析、およびその動的分子間相互作用の解析を試みた。特に、本伝達系はがんの進展に関わっていることが報告されており、本研究の成果は、細胞情報伝達機構の理解だけでなく、新たな抗がん剤の開発につながると考える。
