Outline of Final Research Achievements

We have developed a bio 3D printer technology that creates tissues with complex shapes using spheroids as one unit. By maturing a tubular structure constructed with a bio 3D printer in a bioreactor, we succeeded in producing a small-diameter artificial blood vessel having a mechanical strength (burst pressure test) of 10 to 20 times or more that of human normal blood pressure. Furthermore, we succeeded in a long-term transplantation experiment into immunodeficient mini pigs to which a surgical technique is applied using a small-diameter artificial blood vessel mainly composed of human-derived fibroblasts.

Academic Significance and Societal Importance of the Research Achievements

外来異物を全く含まず細胞のみで3次元的な組織を造る技術を利用し、大動物への長期移植に成功した報告は極めて少ない。虚血性心疾患のみならず従来の人工血管では移植不能な様々な血管不全症例に応用され、多くの人類のhealth careに貢献できる可能性が見いだされた。また本研究のさらなる発展により自己組織での臓器再生・移植モデルが確立されれば、免疫応答、感染の危険性を回避できる次世代の再生治療モデルの確立につながると考えている。
