Outline of Final Research Achievements |
We developed foreign protein production systems using the cold-adapted bacteria, Shewanella livingstonensis Ac10, Pseudoalteromonas nigrifaciens Sq02, and Shewanella vesiculosa HM13, isolated from low-temperature environments as the hosts. We succeeded in constructing a regulatable low-temperature protein expression system using S. livingstonensis Ac10 and a secretory protein expression system using P. nigrifaciens Sq02.

Academic Significance and Societal Importance of the Research Achievements |
低温菌が生産する好冷性酵素は低温で高い活性を有し、低温で使用される食品加工用酵素や洗剤添加用酵素等として有用と考えられている。しかし、好冷性酵素は一般に熱安定性が低く、常温性酵素などと比べて容易に失活する。本研究で開発した低温菌を宿主としたタンパク質生産系は、タンパク質の熱変性が生じにくい低温での外来タンパク質生産を可能にするものであり、熱安定性の低い好冷性酵素の生産などに有用と考えられる。
