Outline of Annual Research Achievements

放射線照射によって生じるDNA二本鎖切断(DSB)は細胞の運命を左右する重篤なDNA損傷の一つである。申請者はDSB修復経路選択性の研究を行う中で、G2期細胞ではNHEJが修復の第一経路であり、NHEJが停滞・遅延した場合に、CtIP/MRE11依存的にHRへと移行することを報告してきた(Shibata, EMBO, 2011; Shibata, Mol Cell, 2014)。一方、他グループの研究から、転写活性領域に生じたDSBはHRにより優先的に修復されることが示されている(Aymard, NSMB, 2014)。そこで本研究では、転写活性領域に生じたDSBがどのような分子機構を経てHRの開始を導くかを明らかにすることを目的とする。
本研究では、転写と共役したHR開始に必要な分子メカニズムの解明を目的とし、申請者がこれまでに発見した転写と連携する新規HR候補因子について、1)DNA損傷部位へのリクルート、2)DNA損傷依存的DNARNAハイブリッドの生成、3)DNA end resectionの過程における分子メカニズムを解析している。これまでの研究により、東京大学・安原崇哲助教との共同研究によって見出された転写共役型HR促進因子であるRAD52/XPGが、G2期細胞における転写共役型DSB修復に関わることを明らかにした(Yasuhara et al.& Shibata, Cell, 2018 *co-corresponding author)。G2期における転写共役型修復を解析する一方で、G1期細胞における役割についても研究の幅を広げ、本研究課題を発展させている。また独自の部位特異的DNA切断アッセイ系を構築し、現在は本アッセイ系を用いて転写共役型のDSB修復機構の解析を行っている。

Strategy for Future Research Activity

G2期のDSB修復機構の解明に加え、現在はG1期におけるDSB修復機構の解明を目指し研究計画を継続する。
