Outline of Annual Research Achievements

ステロイドホルモンは、多細胞生物の発生に重要な役割を担う。ステロイドホルモンが適切なタイミングで機能するには、ステロイドホルモン生合成器官に対して働きかける神経-内分泌メカニズムが重要である。ショウジョウバエを含む昆虫の主要なステロイドホルモンはエクジステロイドと呼ばれ、脱皮と変態の誘導に必須である。これまでに、幼虫期のエクジステロイド生合成器官である前胸腺には、2種類の神経(PTTH産生神経とSE0PG神経)が直接投射し、エクジステロイドの生合成を制御することが知られている。しかし、これら2種類の神経だけでエクジステロイド生合成の精緻な制御がなされているとは考え難い。そこで本研究では、ショウジョウバエの新規前胸腺投射神経の同定を目指した。
様々な種類の神経をマーキングできるトランスジェニック系統のスクリーニングを実施し、新たに同定された2種類の前胸線投射神経に焦点をあて、ショウジョウバエの発育調節に関わる新たな神経-内分泌メカニズムを追究した。それぞれの神経ペプチドを発現するニューロンの機能、およびこれらの神経ペプチドの受容体の機能を遺伝学的手法と生理学的手法を用いて検討し、いずれもショウジョウバエの幼虫から蛹への移行過程でのエクジステロイド生合成に影響することを示した。
特に今年度は、同定されたうち1種の神経と神経連絡をもつ神経群に焦点を当て、エクジステロイド生合成を制御する一連の神経経路を明らかにした。また、これらの研究成果をまとめた論文が本年度に受理された。
