Outline of Annual Research Achievements

ある種の微生物は細胞外膜にシトクロムタンパク質(OM c-Cyts)を局在化させることで、細胞外の電極との電子のやり取り(細胞外電子移動(EET))を可能とする。本研究は、精密電気化学測定によりEET菌内酵素の反応電子数を計測し、反応速度を測定するプラットフォームとなる技術を確立することを目的としており、本年度は光合成細菌を用いた計測を行った。しかし、前年度に試みたモデルEET菌(シュワネラ)の電気化学測定と異なり、数か月の試行錯誤を経てなお光合成細菌に起因する電気シグナルの検出には至らなかった。考察の結果、その原因は光合成細菌の電気化学測定の技術的な難度だと考えられた。そこでオーストラリアへと渡航し、光合成細菌の測定に関する知見を有しているクイーンズランド大学のStefano Freguia准教授のもとで実験を行った。その結果、光合成細菌を利用した電気化学測定系の確立に成功し、代謝と酵素反応速度に対応した電気シグナルを取得することができた。本結果は、モデルEET菌であるシュワネラのみならず、EET能を有する多彩な微生物に対して本酵素反応速度測定手法が利用できることを強く示唆するものである。以上の成果に関し、現在論文を執筆している。
また、前年度の検討により、シュワネラ菌内のOM c-Cytsがヘムの空間配置を反映した極めて強い円偏光二色性シグナルを示すことが明らかになっている。本知見に基づきOM c-Cytsのシグナル強度を定量比較することで、外膜・細胞外固体との相互作用や電子の流れといった生体特異的な環境の変化によりOM c-Cytsの構造が柔軟に変化することが明らかになった。これらの内容はChemical Communications誌、Langmuir誌に原著論文として掲載され、いずれも雑誌カバーとして採択された。
