Outline of Annual Research Achievements

動物は生後、学習を通じて外的環境に順応する。我々は、自転車の運転技術獲得などを通じ、学習早期には注意深い試行錯誤を繰り返し、徐々に運動が自動化・無意識化していくことを日常で経験している。運動学習中にみられる一連の動作の最適化・効率化は、中枢神経系での神経回路の可塑的変化の結果であると考えられる。我々は大脳皮質運動野第5層錐体細胞に着目し、それが受ける興奮性入力を形態学的に観察・定量化した。錐体細胞の樹状突起には棘突起(スパイン)が存在し、主に一つの興奮性シナプスを形成することが知られている。第5層錐体細胞に蛍光蛋白質を発現する遺伝子改変マウスを用い、生体2光子顕微鏡観察を行うと、スパインを視認することが可能である。このマウスを用い、前肢を用いた運動学習課題を与え、その成功率を評価すると同時に2光子顕微鏡下でスパイン動態を観察した。これにより、運動学習に伴って多くの新生スパインが観察された。スパインが受ける興奮性入力源を定めるため、2光子顕微鏡観察したマウスの固定脳薄切標本を作成し、さらに4重免疫組織化学を施した。このpost hoc免疫組織化学により、新生スパインへのシナプス入力前軸索終末が、皮質錐体細胞由来か視床細胞由来かを弁別することが可能となった。この手法により、運動学習の経過とそれによる新規の興奮性シナプス結合の出現との相関を観察したところ、皮質入力を受けるスパインと視床入力を受けるスパインとでは、i) 学習早期での新生頻度 ii) 新生後の長期残存率 が大きく異なることがわかった。このことは、皮質-皮質間連絡と視床-皮質投射結合とが運動学習において異なる役割を果たしていることを示唆する結果であった。さらに電子顕微鏡を使った微細構造の解析や化学遺伝学的検証を加え、現在論文投稿準備中である。
