Outline of Annual Research Achievements

本研究は、骨髄異形成症候群(MDS)の発症機構を遺伝子変異、および変異により誘導されるエピジェネティクス異常の観点から解明し、患者の新規治療開発および予後向上に資することを目的とする。研究計画のうち、スプライシング遺伝子の一つであるZrsr2の条件的ノックアウトマウスを作成し、表現型の解析を行ったほか、同遺伝子欠失がもたらすスプライシング異常をRNAシーケンスで詳細に評価したが、同マウスはわずかな表現型を示すにとどまり、造血器腫瘍を発症しなかった。次に、MDSに高頻度に認められるコヒーシン遺伝子変異の機能解析のため、変異標的の一つであるSTAG2について、ノックアウト(KO)マウスを作成した。Stag2 KOマウスでは、造血細胞の分化異常を生じたが、MDSは発症しなかった。一方、追加でRunx1 KOを行ったStag2/Runx1両遺伝子KOマウスでは、分化障害が増強し、全例がMDSを発症し死亡した。MDS発症の分子機構解明のため、ATACシーケンス、ChIPシーケンス、Hi-C解析などの統合的なエピゲノム解析を行ったところ、Stag2/Runx1 KOにより、エンハンサー・プロモーター間の染色体ループ形成が協調的に障害され、転写一時停止を引き起こしやすい遺伝子群の発現低下に帰結することを示した。
以上より、MDS発症には複数遺伝子変異が重要な役割を果たし得ること、複数遺伝子変異異常により生じる協調的なエピジェネティック制御、特に染色体3次構造の破綻が同疾患発症に重要である可能性を示した。
上記に関連した研究成果について、日本血液学会、日本癌学会などで口頭発表を行ったほか、米国癌学会誌であるCancer Discovery誌に成果を報告した(Ochi et al. Cancer Discov. 2020 Apr 5)。
