Outline of Annual Research Achievements |
病原体から分泌される多種多様なエフェクタータンパク質は宿主の様々な細胞機能を改変し、感染に有利な環境を作り出す。本研究では、世界中の作物生産に甚大な被害を及ぼす植物病原細菌ファイトプラズマに対する新規防除戦略及び園芸品種創出への基盤構築のため、ファイトプラズマのエフェクターの1つ、花の葉化誘導ペプチド「ファイロジェン」による葉化症状の分子機構解明を目指す。 ファイロジェンはファイトプラズマに共通の症状の1つ、花の葉化を引き起こす一方で、ファイトプラズマ属内には、葉化を誘導するファイロジェンが見つかっていない種・系統も数多く存在した。そこで、昨年度決定したファイロジェンの立体構造情報に基づいてファイロジェンの新規探索法を開発した。これにより多くのファイトプラズマからこれまで未同定だったファイロジェンを網羅的に同定した。本成果により、様々なファイトプラズマに配列多様性に富むファイロジェンが広く保存されることを明らかにした。さらに、新しく同定したファイロジェンの機能の多様性解析を行った。その結果、葉化誘導能を失ったファイロジェングループを初めて同定すると共に、葉化誘導能の有無を決定する1つのアミノ酸残基の決定にも至った。本成果は、ファイトプラズマの病原性がファイロジェンという1つのエフェクタータンパク質の、たった1つのアミノ酸によって決定されることを示す成果で、病原体の進化、および植物-病原体相互作用研究の観点から非常に意義深い。 エフェクターの機能分化や多様化は病原体の進化を理解する上で重要であるが、エフェクターの多様化、機能分化の道筋について統合的な理解を試みている事例は少ない。本研究で明らかになったファイロジェンの機能の多様性は、エフェクターを用いた病原体の進化を理解するためのモデルとして有用であると考えられる。
