Outline of Annual Research Achievements

本研究は、1) 社会的行動への動機付けの神経基盤の解明と、2) 幼少期の社会的経験と成年期の適応的行動との関連性の解明を目的として開始された。まず、内的な動機付けを実際の行動出力に変換できる可能性のある神経回路の探索を行い、社会的行動を含め動機付けおよび情動・感情制御に関わる腹側線条体から大脳皮質運動野への多シナプス神経回路が存在することを発見した。その後、動機づけの情報を行動出力に変換できる神経回路メカニズムの同定を試み、腹側線条体から運動野に到達する回路は、線条体―内側黒質―運動性視床核―皮質運動野であることを示した。さらに、新たに発見された腹側線条体―黒質―運動性視床核―皮質運動野という神経回路が、生体内でどのような機能性を有しているのかについて電気生理学的手法を用いて調べた。大脳基底核直接路ニューロンにCreを発現するD1-Creマウスを用いて、Cre依存的な光遺伝学的手法により腹側線条体の直接路ニューロンを活性化すると、大脳皮質運動野の神経活動を変容できることを発見した。これらの結果は、解剖学的知見に加えて、生体内においても腹側線条体から黒質へと繋がる直接路ニューロンの活動変化は大脳皮質運動野の活動に影響を与えることを示唆している。これらの知見は、社会的文脈も含めて、内的動機づけや感情に関わる情報が腹側線条体に由来し、腹側線条体―黒質―運動性視床核―大脳皮質運動野という神経回路を通ることで、行動出力に影響を与えるあるいは修飾できることを示唆している。本研究は脳の中で動機づけの情報が行動出力に変換され得る神経回路メカニズムを示した点で、神経科学分野から心理学、医学まで多岐に渡る分野に影響を与える新知見を提供したと考えられる。
